    「液-液相分離」は特定のタンパク質とRNAが集合し、液滴と呼ばれる膜のない細胞内構造体を可逆的に形成することで、細胞機能を制御する重要な現象である。本研究は、リン酸化酵素mTORC1の下流で制御される液-液相分離を新規に同定し、その分子メカニズムをの解析を行うものである。新たに同定した液‐液相分離制御と疾患との関連を見出すことを目的としている。本年度は、mTORC1の下流で形成が制御される液―液相分離の探索を行った。既知の液‐液相分離について複数の候補を対象に、蛍光免疫染色解析により検証した結果、mTORC1の活性に依存して形成が変化する液‐液相分離を同定に成功した。次に同定した液‐液相分離の制御について、mTORC1からのシグナルに起因した分子メカニズムの同定を行った。申請者の過去の解析から見出したmTORC1の下流でリン酸化を受ける分子群を対象とし、それぞれの遺伝子破壊細胞を調製したところ、同定した液‐液相分離が起こらなくなる分子を同定した。同定した分子のリン酸化修飾の変化が、液‐液相分離に影響を与えるかを検証する実験系の構築を現在は進めている。

    There are two similarities between lifestyle-related diseases and cancer. The first point is that they are in a chronic inflammatory state, and the second point is that abnormal activation of mTORC1 is observed. Based on these similarities, we hypothesized that the transcription factor FOXK1, which is activated downstream of mTORC1, promotes both of lifestyle-related diseases and cancer. We identified that FOXK1 deficient mice were improved fatty liver-related inflammation, fibrosis, and tumorigenesis.

    Myeloid-derived suppressor cells and tumor-associated macrophages accumulate to tumor sites and construct tumor microenvironment, which promote immune suppression and tumor progression. The accumulation of the myeloid cells is known to be dependent on tumor cell-derived chemokine CCL2. The overexpression of CCL2 is clinically observed in various types of tumor. However, it is poorly understood how CCL2 is upregulated in tumor cells. Here, we show that CCL2 transcription is regulated by mammalian target of rapamycin complex 1, which is frequently activated in downstream of PI3K/AKT pathway in tumor cells. The mTORC1-dependent CCL2 expression is mediated by dephosphorylation of transcription factor FOXK1. Blocking mTORC1-FOXK1 axis prevents CCL2-dependent accumulation of myeloid cells and tumor progressions. These results provide a molecular mechanism bridging from tumor initiation to constructing tumor microenvironment.

    Rapamycin, mTORC1 inhibitor, has drug efficacy of anti-cancer. However, effectors of mTORC1 kinase are not fully understood. To explore the effecetors of mTORC1, we carried out phosphoproteomics analysis and newly identified 30 effectors from the total 20000 phosphopeptides. Next, we analyzed FOXK1 as a transcription factor activated by mTORC1, and identified mTORC1-FOXK1-CCL2 signaling pathway.
