アルツハイマー病(AD)などの神経変性疾患の克服は、社会的要請の大きな世界的課題である。そのため、発症分子機構を明らかにして、創薬標的を早急に提示する必要がある。これまでの神経細胞中心に着目した研究の限界が世界規模で懸念されており、脳内環境として神経細胞以外のグリア細胞と神経細胞の相互作用の解析が着目されはじめているが、その複雑性から解析は遅れている。2021年度は、前年度に見いだしたアミロイド病理およびタウ病理をそれぞれの病理に特異的に連動するサイトカイン/ケモカインスクリーニングについて、それぞれのノックアウトマウスとの産仔の解析を行った。まず、アミロイド病理に連動するケモカインA(ChemoA)の役割についての解析では、APP knockin x ChemoA-KOマウスでは、その若齢においてもアミロイドβ量の増加が認められた。分子メカニズムは明らかになっていないが、単一のケモカイン欠損によって病理を変動させる興味深い結果が得られた。次年度以降、さらに高齢のマウスの解析を進め、その分子メカニズムの解明に移行していく。次に、タウ病理に連動するケモカインT(ChemoT)については、交配でトラブルが生じたが、年度終わりに無事に産仔が得られはじめた。新年度から、ChemoA同様に若齢解析から開始する。本研究計画では、申請者らが創出したADモデルマウスを用いて、ケモカインレベルの制御がどのように病理形成に影響するのかを明らかにし、これまでにない新たな創薬標的を提示できる可能性が高く、今後の解析が期待される。

This study aims to validate our hypothesis that the blood-arachnoid barrier possesses the dynamic clearance system for neurotoxic substances in the cerebrospinal fluid and functions as a central nervus system detoxification system. The present study has proposed that α-synuclein, a pathogenic protein, which is known to be and accumulated in the brain of the Parkinson’s disease, would undergo the LDL receptor-related protein-mediated efflux transport from the cerebrospinal fluid. The study has also established the comprehensive proteomics and the LC-MS/MS-based targeted absolute quantification system for extracellular vesicles, pathogenic proteins, peptides, and metabolites to clarify various efflux transport system at the blood-arachnoid barrier.

We established a motivated behavior assessment system to study the effect of Kampo medicine in mice. Using this system, we evaluated the motivated behavior of AppNL-G-F/NL-G-F mice, an Alzheimer's disease model, until they were 39 weeks old. Motivated behavior in some mice decreased. Histological analysis of the AppNL-G-F/NL-G-F mice at 39 weeks old showed that an increased number of cored amyloid plaques in the striatum coincided with decreased motivated behavior. Western blotting analysis of the AppNL-G-F/NL-G-F mice at 9 months old showed decreased dopamine transporter levels in the subcortical tissue, including the striatum. These results suggest a pathway through which cored amyloid plaques damage the dopamine transporter and result in impaired motivated behavior.

Experimental studies of Alzheimer's disease (AD) have largely depended on transgenic (Tg) mice overexpressing amyloid precursor protein (APP). These mice, however, suffer from artificial phenotypes because not only amyloid beta peptide (Abeta) but also other APP fragments are overproduced. To overcome these drawbacks, we generated knockin mice that harbor Swedish and Iberian mutations in the APP gene, and demonstrated that these animals start accumulating Abetaat 6 months and show memory impairment at 18 months without APP overexpression.
We also generated mice containing an additional Arctic mutation; these started depositing Abeta at 2 months and showed memory impairment at 6 months with aggressive neuroinflammation. We provide the research community with these lines in order to distinguish facts from artifacts in APP-Tg mice and to identify pathological mechanisms upstream and downstream of Abeta deposition.

Recently, we found that Ass43 is generated under an age-dependent manner, thereby it might be a good candidate as a marker for an early diagnosis of AD. Herein, we established a high-sensitive and quantitative method to detect Ass43 specifically in the sample from mice and human materials. Finally, we concluded that Ass43 has a potent pathogenicity, and would be an useful candidate for an early diagnosis for AD.
